Cellular fuel gauge may hold the key to restricting cancer growth

Dec. 27, 2012 ? Researchers at McGill University have discovered that a key regulator of energy metabolism in cancer cells known as the AMP-activated protein kinase (AMPK) may play a crucial role in restricting cancer cell growth. AMPK acts as a "fuel gauge" in cells; AMPK is turned on when it senses changes in energy levels, and helps to change metabolism when energy levels are low, such as during exercise or when fasting. The researchers found that AMPK also regulates cancer cell metabolism and can restrict cancer cell growth.

The discovery was made by Russell (Rusty) Jones, an assistant professor at McGill's Goodman Cancer Research Centre and the Department of Physiology, Faculty of Medicine. Jones along with his team is the first to show that AMPK can act as a tumour suppressor in animals. The research will be published December 27 in the journal Cell Metabolism.

"Cancer is a disease in which cells lose their normal restraints on growth and start to divide uncontrollably. But, in order for cells to grow quickly they need enough energy to complete the task," Jones explained. "AMPK acts like the fuel gauge in your car -- it lets the body know when energy levels are low, and stops cell growth until there is enough gas in the tank. We wanted to see if this fuel gauge could affect the development and progression of cancer. We found that mice lacking AMPK developed tumours faster, suggesting that AMPK is important for keeping tumour development in check, at least for some types of cancer." For this study, Jones' team focused specifically on a type of blood cancer known as lymphoma. They discovered that the protein Myc, which is activated in more than half of all cancers, could promote lymphoma more rapidly when mice were deficient for AMPK.

One of the ways cancer cells support their enhanced rate of growth is by changing their metabolism, or how they generate energy. Cancer cells are different from normal cells in our body because they preferentially use sugar to fuel their growth. Jones discovered that AMPK plays a specific role in restricting cancer cells' ability to use sugar to fuel their growth. "For cancer cells with low AMPK levels, their metabolism goes into overdrive," explained Prof. Jones. "They use sugar more efficiently, allowing them to grow faster. These results suggest that turning on AMPK in cancer cells may be one way that we can restrict cancer growth."

Jones' breakthrough builds on his previous discovery that the widely prescribed medication metformin, a common diabetes drug, can restrict tumour cell growth. The results bring promise that common therapeutics that turn on AMPK and alter cellular metabolism, such as metformin, may become novel tools for cancer therapy. Jones and his colleagues at McGill are currently exploring clinical applications based on this research.

How Much Money Can You Make With A Manufactured Home ...

An article I wrote recently, telling the story of my first manufactured home community that I purchased for $400,000 and sold around ten years later for $1,525,000, raised the ire of a self-styled industry ?guru? who contended that such amounts cannot be made with a manufactured home community. Of course, a community we closed on a few weeks ago in Kentucky showed an appraised value of over $1,000,000 more than we paid? so it doesn?t seem that unique to us. But it does raise the question of how much money you can realistically expect to make with a manufactured home community.

How much money do you want to make?

People have different goals. Some people are only concerned with trying to get a higher return on their cash, which is currently getting 1% in a CD. Others are trying to create substantial wealth, far beyond a simple return on their down-payment. It?s a free country, so there?s no law on what you have to make from a manufactured home community purchase.

What you make is totally based on what you buy.

If you are buying communities at 7% cap rates which are 100% occupied, with no room for pushing rents or sub-metering utilities, then it is unlikely that you will create much value other than a 10% or so cash-on-cash return. You?ll run the park for a while, maybe make a small rent increase over time, but you?ll probably sell it back for that same 7% cap rate years later, and have a very small amount of profit going to you at closing. On the other hand, if you buy manufactured home communities at a 10% cap rate, and with significant room to push rents, bill-back utilities and fill lots, and then, after successfully putting all those factors into motion, sell that now 18% cap rate purchase for a 9% cap rate, you will have created a huge amount of value, which you will receive as a cashier?s check at closing. That?s one way that you can create $1 million of value with a manufactured home community.

Don?t forget about buying $1 for 50 cents.

Sometimes you can buy a manufactured home community on the front end, that?s already running well, for less than it?s worth. This is normally a result of a seller who has failed to perform proper accounting on his property, including expensing items that should be capitalized, or not collecting the rent because they are too friendly with their tenants.

Benjamin Franklin summed it up.

Benjamin Franklin once said ?diligence is the mother of good luck?. What I think he meant is that your success or failure is based on how you buy the property on the front end. If your goal is to create a significant amount of capital by buying a property that has great numbers going in, with the possibility of even better performance over time, then you can make a lot of money with a manufactured home community. If, however, you buy a community with lousy numbers and no possibility of improvement, then you will make no money? or even lose your money.

But this is not a ?get rich quick scheme?.

Buying and operating manufactured home communities is far from a ?get rich quick scheme?. But that does not mean it?s not a ?get rich scheme?. The only reason to get involved in this industry is to make money? it?s not a sexy business that sounds good at a cocktail party, nor is it really that much fun. But I think it?s safe to say it?s never a ?quick? business. Holding and operating a community for 5 to 10 years is anything but ?quick?.

As the industry becomes more professional, the opportunities will diminish.

Of course, one of the reasons that there is so much money to be made in the manufactured home community business is that the industry is dominated by moms and pops, who are not the most sophisticated sellers. There are roughly 50,000 manufactured home communities in the U.S., and the majority of these are owned by the original founders or folks who have had them for decades. There are hardly any new communities being built, so this supply is static. As professional investors buy out these moms and pops, the profitability of manufactured home communities? based on going-in cap rates and potential income growth will decrease. This means that there is some sense of urgency, and it is not a given that opportunities will always be as ripe as they are now.

There are two types of business models in this industry.

There are public companies that prefer to buy ?finished? goods that are stable but have a lower return, and others who seek out ?turnaround? opportunities that are non-stabilized, and offer higher returns for those who take the time, effort and risk to stabilize them. There is no reason that these two business models cannot happily co-exist. And also no reason that the lower returns of the one niche should apply to the other. The industry ?guru? who believes that it is impossible to make significant money in this business has been hanging around with the guys at the public companies for too long.

Conclusion

I am proud to make significant returns with manufactured home communities. I firmly believe that there is still plenty of opportunity in this industry. Those who think otherwise either don?t really know what they?re talking about, or don?t have what it takes to be a successful community owner.

Only balanced plan to avoid "cliff" is Obama's: White House

WASHINGTON (Reuters) - The only proposal that avoids the year-end U.S. "fiscal cliff" in a balanced manner is the one President Barack Obama has put forward, White House spokesman Jay Carney said on Monday.

"The only plan that we have seen that achieves the size and the balance that's required for sustainable - for long-term deficit reduction and for putting our economy on a sustainable fiscal path, is the president's," Carney told reporters at a briefing.

Carney had been asked to comment on reports of a proposal from House of Representatives Speaker John Boehner, a Republican, that would allow top tax rates to rise in exchange for cuts to entitlement programs.

Caviar vending machines pop up at L.A. malls

Craving caviar in Los Angeles? There?s a vending machine for that.

Head to three local malls???Westfield Century City, Westfield Topanga and Burbank Town Center ? and you?ll see one of the brand new machines dispensing the prized roe, as well as truffles, escargot and mother-of-pearl spoons.

Prices start at $5, going all the way up to $500, which will get you one ounce of Imperial River Beluga Caviar.

The project is the brainchild of Kelly Stern, who co-owns Beverly Hills Caviar ? a gourmet food supplier located in the tony California community.?

The business plan: Reach customers in wealthy neighborhoods surrounding the malls who may not have convenient access to caviar and expose everyday shoppers to the delicacy, especially younger ones.

?All my friends and family knew about this for a year-and-a-half and no one wanted to tell me that I was crazy, but I could see it in their eyes,? Stern said.

?I did not know what was going to happen when people saw it.?

Stern shopped around the world to find the right kind of vending machine ? it had to keep the caviar cold, have a touch screen, an Internet connection and an attractive display for its gourmet contents. It also had to accept credit cards as well as cash.

She ended up buying parts from seven different countries and putting everything together in Los Angeles. Each machine costs about $85,000???including hardware, software, security and maintenance ? and has about $50,000 worth of inventory inside. The caviar is sold in jars and stays fresh for about a year.

Stern wants to franchise the business to ?every single city in America and the world? when she has more experience with the machines, which she describes as experimental. Stern says no one has ever thought of caviar as a turnkey enterprise, something she wants to change.

?A lot of the people that sell caviar now are old people. They are a dying species and they don?t cater to the young people at all,? Stern said.

?So I?m seeing all the teenagers that stop by as my future assets. They will grow up and become the president of the U.S. or the president of Microsoft and they will think about caviar. They will remember that they went into the mall and saw the machine and that?s where they are going to buy their caviar.?

The project has attracted lots of attention since the machines were unveiled on last week on?Black Friday, prompting Stern to hire a publicist???not to promote the venture, but to manage the curiosity it has sparked.

Industry observers, however, were skeptical whether the business model would be a success.

?It?s interesting, it?s a conversation starter and it?s a marketing tool? (but) I don?t think it?s going to sweep the nation,? said Tim Sanford, editor-in-chief of ?Vending Times,? adding that his staff chuckled when they heard of the concept.

?My guess is that most people who want caviar know how to get it.?

Vending machines often put?much-desired products in high-traffic areas where they can be sold swiftly to people who are there for some other purpose, so the caviar angle sounds more like a way to generate buzz, Sanford said.

But Stern projects she will recoup her money in the next few months, especially with the help of the holidays and awards season in Los Angeles.

?Everybody is talking about it. This is a dream come true,? she said.

Evidence points to police manipulation in SAfrica

JOHANNESBURG (AP) ? South African police may have altered evidence and planted weapons after they shot dead 34 striking miners near Lonmin's Marikana mines in August, according to photographic evidence presented at a commission of inquiry into the killings.

Photographs taken by police the night after the shootings show more weapons by the dead bodies than there were in photographs taken immediately after the violence on Aug. 16. Thousands of miners had gathered at hills in Marikana about 94 kilometers (58 miles) northwest of Johannesburg where 34 miners were shot dead by police and 78 wounded in the worst state violence since the end of apartheid in 1994.

South Africa is conducting a commission of inquiry to look into the parties responsible for 46 deaths, including two policemen and two security guards, during nearly six weeks of strikes at the Lonmin Marikana mines.

Video evidence shown Monday also indicated that some of the slain miners may have been handcuffed.

National police commissioner Riah Phiyega said that the commission has launched an investigation into the discrepancies. She said she was presented with evidence that may have suggested one of the crime scenes had been tampered with nearly two weeks ago.

Human rights lawyer George Bizos said the evidence presented at the inquiry clearly indicates an attempt was made to alter the scene.

"The evidence clearly showed there is at least a strong prima facie case that there has been an attempt to defeat the ends of justice," he said. Bizos, who is representing the Legal Resources Centre and Bench Marks Foundation during the inquiry, called on senior police officers in charge of the scene to present evidence.

Crime scene expert Capt. Apollo Mohlaki, who took the night photographs, was questioned during the inquiry Monday. He admitted his photographs showed more weapons around the bodies than those taken earlier, according to the South Africa Press Association. In one set of photos, a man's mangled dead body lies alone in the daylight, and in a picture taken by electric light after dark, there is a panga (machete) under the man's hand.

Mohlaki said he saw the weapon under the man's arm in the photograph he took, but when looking at the day photograph of the same body he said of the weapon: "It is not appearing, I don't see it."

Dali Mpofu, the attorney for the Lonmin miners, entered a video as evidence that showed miners that seemed to be handcuffed. When asked if he saw if any of the dead miners' hands had been bound, Mohlaki said he had not.

"If I am looking at the video there is a person, handcuffed possibly, but on the day I did not observe that," Mohlaki said.

The representative for the police, Ishmael Semenya, had suggested the week before that the integrity of the crime scene could have been compromised by the presence of paramedics, according to SAPA.

"We will hear evidence that paramedics asked that weapons be removed so they could do their work," said Semenya.

On Tuesday, the commission saw photographs of the security guards' bodies, who were among those killed before the Aug. 16 shootings. The photographs showed one of the guards with his tongue cut out while the legs of the other security official were visible in the charred remains of a security vehicle torched by the protesters, according to SAPA. Another photograph showed a group of marchers fleeing with a shotgun stolen from the security guards, it said.

The inquiry began last month and is expected to continue for four months, investigating the roles played by police, miners, unions and Lonmin in the August deaths. Strikes at Lonmin spread to other mining sectors last month, and a deal reached with Lonmin miners that saw workers getting a pay increase of up to 22 percent has workers at other mines demanding similar deals.

Gold Fields said Tuesday that thousands of employees returned to work at the company's South Africa KDC East mine ending a 23-day strike. All three of its mines in South Africa are now back in production, it said. Gold Fields, which is one of the world's top producers of gold, had dismissed 8,100 employees in October for participating in illegal strikes, however it says a deal reached with the National Union of Mineworkers saw a vast majority of those workers reinstated.

The NUM said Tuesday that the 12,000 miners fired from platinum mining company Anglo American Platinum in Rustenburg for staging illegal strikes since September had still not returned to work, and negotiations were underway.

How pulsars slow down with age

ScienceDaily (Oct. 8, 2012) ? Researchers at the University of Southampton have developed a model which explains how the spin of a pulsar slows down as the star gets older.

A pulsar is a highly magnetised rotating neutron star which was formed from the remains of a supernova -- an explosion which happens after a massive star runs out of nuclear fuel. A pulsar emits a rotating beam of electromagnetic radiation, rather like that of a lighthouse. This beam can be detected by powerful telescopes when it points towards and sweeps past Earth.

Pulsars rotate at very stable speeds, but slow down as they emit radiation and lose their energy. Professor Nils Andersson and Dr Wynn Ho, from the University of Southampton, have now found a way to predict how this 'slowing' process will develop in individual pulsars.1

Nils Andersson comments: "A pulsar's spin rate can be a very precise measurement of time which rivals the best atomic clocks, but in the end it will slow down. Until now, the nature of this slowing hasn't been well understood, despite 40 years of research. However, our model will open the door on this process -- extending our knowledge of how pulsars' operate and helping to predict how they will behave in the future."

As a hot pulsar cools, its interior increasingly begins to turn superfluid -- a state of matter which behaves like a fluid, but without a fluid's friction or 'viscosity'. It is this change of state which gradually affects the way that the star's rotation slows down.

"The effect on the star's rotation is like a figure skater extending their arms to slow their spin," says Wynn Ho. "Our model can explain the observed behaviour of young pulsars, such as the 958-year-old pulsar in the Crab nebula, which spins at 33 times a second."

The Southampton scientist's findings have important implications for the next generation of radio telescopes being developed by large international collaborations, like the Square Kilometre Array (SKA) and the Low Frequency Array (LOFAR), of which Southampton is a UK partner university. The discovery and monitoring of many more pulsars is one of the key scientific goals of these projects. Professor Andersson and Dr Ho's mathematical model can be used in conjunction with these observations to predict how a pulsar's rotation will change over time and enable scientists to peer inside these stars and explore their exotic composition.

"Our results provide a new method of linking the study of distant astronomical objects to laboratory work on Earth in both high-energy and low-temperature physics," says Professor Andersson. "It is an exciting example of interdisciplinary science."

1 Pulsars are rotating neutron stars that are seen to slow down, and the spin-down rate is thought to be due to magnetic dipole radiation. This leads to a prediction for the braking index n, which is a combination of the spin period and its first and second time derivatives. However, all observed values of n are below the predicted value of 3. We provide a simple model that can explain the rotational evolution of young pulsars, including the n=2.51 of the 958-year-old pulsar in the Crab nebula. The model is based on a decrease in the effective moment of inertia due to an increase in the fraction of the stellar core that becomes superfluid as the star cools through neutrino emission. The results suggest that future large radio monitoring campaigns of pulsars will yield measurements of the neutron star mass, nuclear equation of state, and superfluid properties.

ShareMyPlaylists Gets A Much Needed Android Reboot

NEW YORK ? If you're keeping a list of journalists who have claimed for years to be Pulitzer Prize nominees without the inconvenience of actually being nominated for a Pulitzer Prize, add one more name: Charles Gasparino, the pugnacious senior Wall Street correspondent for Fox Business Network.

After the outing in this space of non-nominees Jonah Goldberg (conservative columnist,?Pulitzer nomination claimed on the book jacket) and Betty Liu (Bloomberg Television morning anchor,?Pulitzer nomination claimed in ads on commuter trains), a reader wrote to ask, when exactly was Charlie Gasparino a Pulitzer nominee?

Let's see. Until Tuesday afternoon, Gasparino's bio from Fox Business said he was "nominated for the Pulitzer Prize in beat reporting" in 1992, when he was?a reporter for The Wall Street Journal. The same claim is made by his agents at the?HarperCollins Speakers Bureau, and on the website of his publisher,?Simon and Schuster.?In a promotional video in 2008 for CNBC, his former employer,?Gasparino declares, "I am: a writer, son of an ironworker, son of New York, Golden Gloves prospect, a Pulitzer Prize nominee..." (CNBC is owned by NBCUniversal, which is a partner with Microsoft in msnbc.com.)

Next step: Checking the official list of Pulitzer winners and nominated finalists for 1992. Though there are only three nominees, known as nominated finalists, in each Pulitzer category each year, there are more than 2,000 submissions. The winner in beat reporting in 1992 was Gretchen Morgenson, one of Gasparino's competitors on the Wall Street beat for The New York Times. The two other nominees were Patrick Healy of The Boston Globe for education reporting, and Jack Kelley of USA Today for reporting on terrorism. (Kelley turned out to have made up information in his articles, but that's a topic for another day.) No Gasparino.

Fox retracts the claim
When asked on Tuesday in which year he was nominated, former boxer Gasparino jabbed back in a one-line email: "I was nominated by the wsj sir."

But the news organizations don't choose the Pulitzer nominees, any more than the record studios choose Grammy nominees. By Gasparino's reckoning, thousands of journalists each year could sell books and earn speaking fees by calling themselves "Pulitzer nominees."

Later Tuesday, Fox changed its online bio of Gasparino, keeping the P word but dropping any claim to a nomination, saying instead that his work "was submitted for the Pulitzer."

A Fox spokeswoman also sent over a statement:

"The Wall Street Journal submitted Charlie Gasparino's reporting of Wall Street research scandals to the Pulitzer Board in 2002," said the statement from Kevin Magee, executive vice president of Fox Business Network. "While Fox Business never claimed he was a finalist for the award, we've clarified his bio to reflect the submission as opposed to a nomination."

Neither Fox nor Gasparino would answer the question: Why include a "submission" in a bio at all if it didn't make the finals?

Pulitzers "discourage" such puffery
It's not uncommon for Pulitzer entrants to make false claims to be nominees. If all Pulitzer entrants could be called nominees, any publisher could give all its authors and journalists that honorific by submitting an entry form and a check for $50. (And some publishers do seem to play that game.)

As the Pulitzer board's online list of frequently asked questions explains politely, the finalists and the nominees are the same three people in each category: "Work that has been submitted for Prize consideration but not chosen as either a nominated finalist or a winner is termed an entry or submission. ... We discourage someone saying he or she was 'nominated' for a Pulitzer simply because an entry was sent to us."

Caveat emptor
The old journalism motto was, If you're mother says she loves you, check it out.

The new motto: If a journalist or author uses the words "Pulitzer nominee" or "nominated for a Pulitzer Prize," check it out.?The searchable list of winners and nominees is on the Pulitzer site at Columbia University.

Pond scum gets top billing as future fuel source

Algae, known to most of us as pond scum, is the most promising sustainable source of energy to meet the growing demand for juice to power everything from cars to factories, according to a major technical organization.?

?There is not an infinite amount of oil in the Earth and we are using it quite rapidly,? William Kassebaum, a senior member with the Institute of Electrical and Electronic Engineers, the world?s largest technical professional organization, told me today.

While there?s plenty of debate over when the oil will run dry, ?eventually we will need to find other sources to replace these hydrocarbon fuels,? Kassebaum, who is also CEO of Algaeon Inc., an Indiana-based algae producer.

In a statement issued Tuesday, the IEEE endorsed algae as ?the best candidate to provide a sustainable energy source to meet increasing global demands.?

The potential of algae-based biofuel has long been known, but not seriously considered largely because making a barrel of the stuff was?too expensive compared to traditional fossil fuels.?

?Everyone understands that you have to be able to ship a product at a competitive price with fossil fuels,? Kassebaum said.?

When the seminal research on algae-based biofuels was conducted in the 1970s, the price of a barrel of oil was around $20. Today, oil is around $80 with recent spikes over $100 a barrel, a price range where algae fuel producers believe than can compete.

Doing so, Kassebaum noted, will take major capital investment and perhaps government support to enable the industry to produce commercially viable quantities, but in the coming years he believes algae ?will become part of the oil industry.?

The industry will still need?to confront concerns such as water supplies ?? perhaps by producing algae in parts of the country where water supplies are abundant, such as the Midwest, where Algaeon is located.

The Midwest, Kassebaum noted, is already?rich with corn, much of it grown for ethanol.?

In the IEEE statement, he noted that ?An acre of corn can be used to generate 300 gallons of ethanol per year, while an acre of algae can produce 6,000 to 10,000 gallons of light sweet crude oil annually.?

Put that way, perhaps algae really is worthy of the endorsement.

At The Rim: Disabled Couple Compile Encyclopedia for the Deaf

The first encyclopedic dictionary for the country's 20 million deaf and hear-impaired people was published early June, its compilers, a disabled couple in east Anhui province, told Xinhua Thursday.

The 200,000-word reference book embodies more than 1,000 entries about medicine, education, psychology, sociology, rehabilitation, linguistics, law, history and culture concerning the life of deaf people, according to the couple living in the provincial capital of Hefei.

"We first collected and sorted out relevant material by ourselves, and then consulted a number of experts across the country and invited them to make revisions to the book," said compiler Shi Li, a deaf woman who serves as deputy head of the provincial association of deaf people.

The exhaustive work, priced at 78 yuan (12 U.S. dollars), took Shi and her lame husband, Cui Jiping, six years and cost over a million yuan to compile.

"(Before reading the book,) I never imagined that there were records about the culture of deaf people dating back as early as the Warring States period (475 B.C. to 221 B.C.), and there were laws specially made to protect our rights," said Wang Hongqing, a deaf man in his 50s.

"The book shows that we're not alone, since it presents so many exchanges between the deaf across the world," Wang said.

The encyclopedia provides the deaf with a rational perspective on themselves and can help ordinary people know more about the group, which is significant in building a discrimination-free society, a senior official with the China Disabled People Union said in a congratulatory letter sent to the couple.

Shi said the idea to publish an encyclopedia for the deaf had long fermented in her mind.

"Many deaf people I met during my work raised the question that 'where can I get the information about my condition?' Some experts also mentioned the necessity of having a comprehensive book on the issue," said Shi, who went deaf after being improperly injected with an antibiotic drug at the age of three.

The compiling of the book was funded by the couple's company, which specializes in auxiliary facilities for the disabled.

In the past five years, the technology company, of which 70 percent of its employees are disabled, has invented, manufactured and sold dozens of products, such as the wireless doorbell alerting system for the deaf.

The 43-year-old Cui said the research and development team of the company is working on developing a global network that will allow disabled people to fully enjoy information and video services.

"In the next step, I've planned to open a museum for the deaf," he said.
